Towards Standardized Extraction from Formalin-Fixed Paraffin-Embedded Tissue:
- Aims to develop a reliable method for DNA extraction from FFPE tissues for public health decision-making, despite challenges from formalin fixation and processing variations.
- Plan involves 90 labs contributing tissue blocks and using standardized protocols alongside one optional method. Results to assess DNA yield and HPV detection in integrated study later 2010.
Study Comparing Commercially Available HPV Genotyping Assays:
- Part of a WHO-LabNet collaboration.
- Assessed 3 kits (Linear Array, INNO-LiPA Genotyping Extra, RhodamiR-HL) on sensitivity, specificity, reproducibility, time, and ease of use.
- Used four sample series: control plasmids, single type plasmids, mixed type plasmids, and real-world extracts (epidermoid cell lines, CIN tissue FFPE, cervical scrapes transport media).
- Goal: Identify the best kit for large-scale use in HPV genotyping programs. Study initiated Oct 2010, results expected Jan 2011.
An Update: HPV LabNet International Collaborative Studies
- Standardized HPV Antibody Threshold: Collaboration with NIBSC to establish a primary international standard for anti-HPV 18 antibodies (IS) via a proficiency study using freeze-dried serum.
- HPV DNA Proficiency Panel: Worldwide assessment of genotyping methods, e.g., 4th Panel (2010) evaluated suitability of E1-targeted PapilloCheck.
- International Standard for HPV 16 E6/E7 mRNA DNA: Development by US CDC GRL to be proposed as a reference standard.
HPV LabNet Regional Reference Laboratory Activities: A Snapshot
- Western Pacific Region (Swiden, Australia, Canada, France, Germany, Norway, Slovenia):
  - Japan: Prevalence survey (52, 16, 58 dominant), developing high-throughput typing, participating in DNA Proficiency Panel.
  - Australia: HPV surveillance both pre-vaccination and post-vaccination studies, research into other HPV-related cancers, participates in LabNet activities and meeting reports.
  - France: National cervical cancer screening genotyping program (Beta-papillomavirus research).
  - Germany: National Reference Center development, testing new assays (e.g., Nuclisens Easy Q).
  - Canada/Sweden/Norway: HPV vaccine surveillance programs (urine testing ongoing/study) using molecular assays.
Meeting Reports/Updates
- WHO Meeting on HPV Vaccine Coverage and Impact Monitoring: Discussed impact monitoring, significance of harmonized cancer registries, limitations of testing HPV prevalence in sexually active young people as a measure of effectiveness, and importance of reporting to registries. Meeting Oct 2010.
- International Papillomavirus Symposium on HPV in Vaccinology and Surveillance: Focused on reviewing LabNet activities in GRL/RRLs, discussing expertise of National Reference Laboratories (NRLs), collaborations, and strategies (e.g., HPV QA programs in Scotland, South Africa, and overall activities of GRL). Conference July 2010.
