Abstract: Mitochondria-associated caspase-recruiting structure (MACS), formed by the rearrangement of outer membrane proteins and mitochondrial outer membrane translocation complex (MOMT) pore during apoptosis, serves as an intracellular signaling platform that directly recruits caspase-9 into the mitochondrial intermembrane space (IMS) for subsequent activation of caspase-3/-7. Here, we report that, distinct from its role in apoptosis, the disruption of MACS formation caused by Mdivi-1 (mitochondrial division inhibitor 1) inhibits G2/M cell cycle progression and triggers mitotic catastrophe in non-small cell lung cancer (NSCLC) cells. Our results demonstrate that Mdivi-1 treatment led to depletion of phospho-RDSII and failure in nuclear envelope breakdown (NEB) in a ROS-dependent manner. Mechanistically, Mdivi-1-mediated mitochondrial hyperfusion and MACS breakdown cooperatively promoted the accumulation of USP19 at IMN, which deubiquitinates and stabilizes L1CAM in the nucleus to stimulate the activation of AKT/GSK3B/BATF3/p21 pathway, thus promoting S phase delay and NEB inhibition with subsequent mitotic catastrophe formation. Furthermore, aberrant expression of USP19 and L1CAM and MACS breakdown are associated with poor survival of NSCLC patients. Collectively, our study uncovers a novel Mdivi-1/p53-independent pro-apoptotic activity in mitotic catastrophe triggered by ROS-mediated hyperfused mitochondrial network and subsequent aberrant nuclear-cytoplasmic shuttling of USP19/L1CAM axis in NSCLC cancer cells, providing a novel rationale for targeting at hyperfused mitochondria through Mdivi-1 to block cell cycle progression and trigger selective anticancer therapy.
